This is an unformal way of speaking and is very common in the speech of children from London. It basically means 'Exactly' , 'For real' or 'Really?'

To be serious, honest, or truthful about something.

Ellen told me she was gay, I responded by saying “On word?” then she said “Yes.” Fred said he got a tattoo, I didn’t believe him so I said “On word?” And then he showed it to me. Stacy’s boyfriend told her he loved her. She didn’t believe him so she said “On word, you love me?” He replied with “On word.”
